Lgr5 is the R-spondin receptor. Lgr5 resides in Wnt receptor complexes and mediates signaling of the R-spondin Wnt agonists (33), explaining the unique dependence of Lgr5 stem cells of various epithelia on R-spondins in vivo and in vitro. Two other Wnt target genes, RNF43 and ZNRF3, encode stem cell-specific E3 ligases that downregulate Wnt

Lgr5, a member of the G protein-coupled receptor family, is a Wnt target in the gastrointestinal and integumentary systems. Lgr5 is thought to function as a potentiator of Wnt/β-catenin signaling in epithelial cells. Upon deletion of Lgr5 in B-ALL cells, however, we observed massive accumulation of nuclear β-catenin and increased expression of β-catenin target genes, which suggests that Lgr5 acts as an essential negative regulator of β-catenin in the B-cell context. Wnt signaling regulates Lgr5 expression. In the gastrointestinal and integumentary systems, Wnt signaling controls Lgr5 expression (Jaks et al. 2008; Ootani et al.
LGR5, CEACAM6 and Wnt pathways are suppressed by shCD151 in HT29 and HCT116 cells in vitro and in vivo Western blot and qRT-PCR demonstrated significantly decreased expression levels of LGR5, CEACAM6, β-catenin, Wnt3a and Wnt5a in cultured HT29 and HCT116 cells after CD151 silencing at the protein and mRNA levels, respectively ( Figure Figure8 8 ).
LGR5 is a known WNT pathway target gene and marker of intestinal stem cells. The LGR5+ stem cells are located in the crypt base and capable of The receptor LGR4 promotes Wnt signaling in response to the binding of R-spondin ligands (RSPOs) both by inhibiting the activities of the E3 ligases RNF43 and ZNRF3 and by directly stimulating the Wnt signalosome through the scaffold protein IQGAP1. It is thought that LGR5 stimulates Wnt signaling similarly to LGR4 although LGR4 and LGR5 are not functionally equivalent in vivo.
Lgr5 homologues are facultative Wnt receptor components that mediate Wnt signal enhancement by soluble R-spondin proteins. These results will guide future studies towards the application of R-spondins for regenerative purposes of tissues expressing Lgr5 homologues.
We find that conditional deletion of both genes in the mouse gut impairs Wnt target gene expression and results in the rapid demise of intestinal crypts, thus phenocopying Wnt pathway inhibition. Mass spectrometry demonstrates that Lgr4 and Lgr5 associate with the Frizzled/Lrp Cadigan KM, Waterman ML (2012) TCF/LEFs and Wnt signaling in the nucleus.
Lgr5 + stem cells occur in other epithelia such as the one of the colon, stomach, hair follicles ( Alonso and Fuchs, 2003 ), and mammary glands.

TGFβ1 additive activates LGR5, CEACAM6 and Wnt signaling after CD151 silencing. A and B. Western blot and qRT-PCR showed TGFβ1, LGR5, CEACAM6, Wnt3a and Wnt5a were remarkably upregulated at both protein and mRNA levels after treatment with TGFβ1 at 0.05, 0.10 and 0.50 ng/ml, respectively, on sh-CD151 transfected HT29 cells (sh-CD151+TGFβ1 group versus sh-CD151 group). The treatment experiment was performed in an intracranial orthotopic xenograft model by knockdown of LGR5 or by using the Wnt/β-catenin pathway inhibitor Wnt-C59. LGR5 expression was determined in 268 glioma specimens by immunohistochemistry.
